FAQ: Auto Exposure Settings

Question

Why when saving the settings and opening the software again the saved settings are only active for the first image? i.e. the camera gain is switched back on 8 automatically.

Answer

If you are using automatic exposure and the the exposure algorithm is anything other than 'Simple Linear' then the auto exposure algorithm will adjust both the exposure time and the gain. The idea is that on automatic it will choose the best combination of gain and shutter time to maximise image quality and minimise fluor fading.If you wish to keep the gain fixed

  1. open the device editor and select your camera from the list of devices.
  2. click on Setup
  3. Choose 'Peak Value' from the 'Mode" popup
  4. Choose 'Auto Shutter' from the 'Threshold' popup. Our recommendation for general FISH imaging is to use 'Full Auto 0.5'
